چکیده

Abstract Background Despite the improvement of relapse-free survival mediated by anti-angiogenic drugs like sunitinib (Sutent®), or combinations with immunotherapy, metastatic clear cell Renal Cell Carcinoma (mccRCC) remain incurable. Hence, new relevant treatments are urgently needed. The VEGFs coreceptors, Neuropilins 1, 2 (NRP1, 2) expressed on several tumor cells including ccRCC. We analyzed role VEGFs/NRPs signaling in ccRCC aggressiveness and evaluated relevance to target this pathway. Methods correlated NRP1, levels patients’ using online available data base. Human mouse were knocked-out for NRP1 NRP2 genes a CRISPR/Cas9 method. number metabolically active was XTT assays. Experimental generated immuno-competent/deficient mice. effects competitive inhibitor 2, NRPa-308, tested vitro vivo above-mentioned tests experimental NRPa-308 docking performed both NRPs. Results Knock-out inhibited metabolism migration stimulated expression VEGFC, respectively. presented higher affinity than NRP1. It decreased migration/invasion more efficiently commercially NRP EG00229. robust inhibition growth immunocompetent immunodeficient Such associated pro-tumoral factors. Analysis TCGA database showed that pathway, pathway correlates only patients. Conclusions Our study strongly suggests inhibiting NRPs is treatment mccRCC patients therapeutic impasses represents hit.

Differential expression of neuropilin-1 and neuropilin-2 in arteries and veins

Neuropilin-1 (np1) and neuropilin-2 (np2) are receptors for class-3 semaphorins and for several isoforms of VEGF. We have cloned and characterized two chick isoforms of np2 cDNA. Expression patterns of np1, np2, and ephrin-B2 were compared in the developing vascular system of 24-72 h old chick embryos. We show for the first time that np2 is expressed in blood vessels in vivo from the earliest s...

Metabolic stress induces the lysosomal degradation of neuropilin-1 but not neuropilin-2.

The neuropilins-1 and -2 (NRP1 and NRP2) function as receptors for both the semaphorins and vascular endothelial growth factor. In addition to their contribution to the development of the nervous system, NRP1 and NRP2 have been implicated in angiogenesis and tumor progression. Given their importance to cancer and endothelial biology and their potential as therapeutic targets, an important issue...

Neuropilin-1 and neuropilin-2 act as coreceptors, potentiating proangiogenic activity.

Neuropilin-1 and -2 (NRP1 and NRP2) are the transmembrane glycoproteins interacting with 2 types of ligands: class III semaphorins and several members of the VEGF family, the main regulators of blood and lymphatic vessel growth. We show here that both NRP1 and NRP2 can also bind hepatocyte growth factor (HGF). HGF is a pleiotropic cytokine and potent proangiogenic molecule that acts on its targ...

Neuropilin-1 suppresses tumorigenic properties in a human pancreatic adenocarcinoma cell line lacking neuropilin-1 coreceptors.

Neuropilin-1 (NRP-1) was first described as a coreceptor implicated in neuronal guidance that bound members of the semaphorin/collapsin family. NRP-1 is also expressed in endothelial cells and is believed to promote angiogenesis by acting as a coreceptor with vascular endothelial growth factor (VEGF) receptor 2. Recent studies suggest that NRP-1 can function through both a VEGF-dependent and VE...

Sunitinib in Metastatic Renal Cell Carcinoma: The Pharmacological Basis of the Alternative 2/1 Schedule

Sunitinib is an inhibitor of platelet-derived growth factors receptor (PDGFR) and vascular endothelial growth factor receptor-2 (VEGFR2) tyrosine-kinase activities, and it is registered for the treatment of metastatic renal cell carcinoma (mRCC). Indeed, sunitinib 50mg/day for 28 days every 6 weeks is effective against mRCC and induces significant improvements in progression-free and overall su...
